Drill away.
I was an electriction for about 15 years. Got out of it last July.
Just try to keep the holes in the center of the stud. 4x6 beams, 2x4, 2x6 stacked can be hard to keep the holes in the center so you will need to use nail plates where the out side edge of the hole is closer than 1 1/4 inches to the outside of the stud. I have drilled hundreds of thousands of these and never had a problem. Just try to use good judgement. Keep the holes no larger than 7/8 if you can help it. And spread the holes evenly up and down don't bunch the holes all in one spot.
You can drill a G.L Beam if you get permission from the architect. And you can drill a G.L at the ends of the beam. Where the beam and wall meet draw a 45 deg. line up to the top of the beam. You can drill anywhere above that line.
